Has anyone had success with an in-house von Willbrand Factor collagen binding assay (Type I)?

Currently using vWF:CBA Collagen I 96/well kit in a research lab. They are pretty expensive, and sometimes have issues of unavailability. Has anyone had success with a recipe for an in-house assay?

Can multiple freeze-thaw cycles be performed on a hydrogel (PVA/PVP) for crosslinking without changing the mechanical properties?

The polymer goes through 6 freeze-thaw cycles (21 hours at -19C and 3 hours at 25C). Does anyone know, if I perform 2 or 3 cycles in a day, would the polymer be the same?
