How can i do chlorophyll determination (anatomical studies) on dragon plant cuttings without using a chemical?

I have been searching for a different way of determining the chlorophyll content in dragon plant (pitaya) cuttings without the use of Dimethyl Sulphoxide (DMSO) or any other chemical.
