Using FITC conjugated Annexin-V for apoptosis detection. Why in some protocol, they do not wash the cells after staining ?

Could someone kindly explain to me why some protocol do not wash the cells after staining with FITC-Annexin V and PI, to get rid of unbound annexin V/PI ?

I am going to express CD36 in HEK cells, which plasmid I should use and do you recommend specific company to buy the plasmid with the construct ?

I am going to express Human CD36 on a mammalian cells, so I wonder if there's a company that I could buy the construct in a plasmid, and if they also provide positive and negative controls. Regards

Why THP1 cells only express 12% CD36 and 20% CD14 and 25% CD45, when I have differentiated them using PMA it showed only increase in CD14 to 30%?

as Cell line shouldn't the whole cells express the same receptors, I wonder why it's really small percentage of cells only express that, I have repeated the experiment many times and always get...
