How to quantify biofilm biomass, maximum thickness, and surface to volume ratio from CLSI image, using Image J??

I tried using COMSTAT plugin in Image J, but it is unable to analyse .czi format. Can anyone help?? Or is there any other way to analyse biomass, maximum thickness, and surface to volume ratio...
