Say a certain bacteria culture gives a certain OD value at 600 nm (say 0.3 OD) after overnight culture. I need the same bacteria suspended in PBS with the same OD everytime (say 0.1 but not 0.3)...

Can someone tell me how to obtain CD spectra till 190 nm when HT values are increasing fairly over 600 at 195 nm with buffer itself?
