Is it possible to express transgene in this lentiviral vector by cloning the cDNA after the EF1alpha promoter? The EF1a promoter which is driving the Mcherry expression and also the PuroR genes.

I want to clone this cDNA and over express them in MSCs can it be possible to amplify the CDS with gene specific primers? or is there any other procedure to amplify from tissue specific whole RNA?
