How to induce lipopolysaccharide induced inflammatory damage in HaCaT cells?

i have tried various doses from reported literature such as 12 mcg/mL for 12 h. i have moved to 12-48 mcg/ml for 24 h. No effect on cell viability via MTT is seen. I am using LPS from E.coli.

Is there any standardized protocol for semi-dry transfer of 12% gel on pVDF membrane (biorad) using transblot system??
