Has anyone performed intracellular stainon on Human Umbilical Vein Endothelial Cells on a 96-well plate? Issues regarding washing excess buffer/mAb?

Looking to perform intracellular staining on HUVECs in 96-well plates, but need to wash off excess permeabilization buffer and antibodies, but don't want to take them out of the wells if possible.

Has anybody used phospho-antibodies for phosphorylation status of proteins/enzymes by intracellular flow cytometry?

Looking to use phospho-antibody (Ser177) for p-eNOS staining by flow cytometry in human umbilical vein endothelial cells.
