Do you scrape and count the PBMC-derived macrophages prior to treating them, in order to plate the same number of macrophages/well for ELISA?

Or can I assume that starting with the same number of PBMCs in all the wells, I will end up with the same number of macrophages in all the wells? (similar enough to not bother with scraping)
