Can Anyone help on trouble to detect Ebf2 protein by WB using abcam (ab94791) antibody?

Protein was collected from C3H10T1/2 cell by using RIPA lysis byffer. Different time point and different conc. of Ab were checked but no luck.

Can anyone suggest how I can visualize number of mitochondria in one cell by con-focal microscope?

I thought of cell surface staining (cell membrane) plus mitotracker and DAPI. Is this possible?and then which cell membrane staining will be feasible for this?
