How can I label single guided RNA (sgRNA) with Alexa 488 dye and quantum dots (QDs)? I will appreciate any paper recommended or detailed protocols.

How can I clone dCas9 from one vector into another vector suitable for its purification?

I purchased pdCas9 from addgene and want to clone and express it in pET22b vector. Please, I need a detailed explanation. Also, where can I get purified dCas9 for purchase? Thanks. O.J
