Can someone advise on Arginase-1 inhibitor?

I spent a long time to look for arginase-1 specific inhibitor, but could not find. Is there anyone who knows this kind of drugs? It seems most drugs target both arginise-1 and -2.

To stimulate T lymphocyte, people use either soluble anti-CD3 or immobilized anti-CD3. May I ask what is the difference between them?

It seems that immobilized anti-CD3 has stronger stimulant effect.

To stain for cytokines in lymphocytes (e.g. IFN-g), are simultaneous stimulation and Golgi block essential?

I want to use an antibody to stain IFN-g in lymphocyte. After initial activation for 2 days, is it essential to add an extra stimulus (e.g. anti-CD3e) when I add in Golgi stop?
