I am about to run mice brain samples in the HPLC. I have optimised the standards however some methods are spiking their samples. Why do I need to do this?

I have mice brain tissue to perform LC/MS for Tryptophan Metabolites. I have also been told to look for inflammatory cytokines. Does anyone have experience with this or recommendations?
