Is it right to leave tissue biopsy in the freezer 24 hrs with or with out media then estimate the cancer stem cells after 24 hrs.?

we receive tissue at 5 pm for cancer stem cell estimation 2nd day 10 am , is it better to leave the tissue at 4 C or -20C ?

Herbal extract has antitumor activity on malignant tissues, on another tissues has not the same effect ,Why?

I tested an extract on cancer stem cells , some times has good effect ( 60% ) some times 2% on another tissue, and some times has no effect ? where is the fault ? OR/ tissues are differently...
