Is it right to leave tissue biopsy in the freezer 24 hrs with or with out media then estimate the cancer stem cells after 24 hrs.?

we receive tissue at 5 pm for cancer stem cell estimation 2nd day 10 am , is it better to leave the tissue at 4 C or -20C ?
