I want to present HCMV peptides to monocytes and then see the T cell activation. I want to know monocyte and Lymphocyte concentration?

My biggest concern is to figure out how many PBMC or adherent cells I need to present my HCMV peptides? Further PBMC (for Lymphocyte) require for T cell activation and cytokine production.
