Can anyone suggest a reliable protocol for transdifferentiation of HUMAN fibroblasts into skeletal muscle progenitors cells and/or muscle fibers?

I´ve found protocols for other species and I´m still looking for human fibroblasts. I don´t want to reprogram the cells to iPS, the goal is differentiate directly to muscle.

How do I optimize myofibril isolation from skeletal and cardiac muscle cells?

I often isolate myofibrils using a homogenization protocol, but it is very common to get bundles of myofibrils (specially from cardiac cells). I wonder why and how I could optimize this isolation...
