Can I get valuable information from the alkali solution resulted from the alkali treatment of plant fiber using UV/Vis or any other methods?

I treated the fiber with 5% NaHO, @ 80, for an hour, twice. The solution get black in the first and light blue in the second treatment.

During alkali treatment of plant fiber the solution gets blacken in the 1st treatment & light blue in the 2nd . Could some one give me justification?

5 % NaOH (W/V), sample solution proportion of 1:60, @ 80 degree delicious for an hour.
