How do you adjust standard settings and FSC gate for detection of apoptosis using flow cytometry with annexing V/FITC staining?

I am inducing apoptosis in HCT-116 cells with Doxorubicin and am not sure how to adjust the initial setting on the flow cytometer.

There might be issues with using Trypsin to remove adherent HCT-116 cells and staining with Annexin V to ananlyze apoptosis using flow cytometry.
