Can any one tell me what to do If in karyotype anlysis from leaf I get too much oil in my slide also cell does not get swollen as well?

Kindly tell me what to do karyotype from leaf sample of Santalum album. The cell does not goes into swollen condition and chromosome also not get swollen and cell are not get swollen also.
