Regarding Indirect Elisa with recombinant proteins?

I'm using purified recombinant proteins to coat for Elisa. As a negative control, I used only the vector without the insert. Reactivity is seen also in that vector. How to troubleshoot this?

To set the Annealing Temperature,

Hi everyone, If two primers have melting temperature of (forward ) 69.33°C and (reverse) 64.03°C, what would be the annealing temperature ?? Help me in finding the PCR protocol. Kindly reply
