When conducting an MTT assay to evaluate synergism between two drugs, do I add the same amount of the drugs as in their combination?

For example, if I am adding a 100uL of each drug in the single treatment wells, do I add 200 uL of them combined in the co-treatment wells or just add 100 of their combination?
