This question has been answered a number of times already on this platform.
You do not measure zeta potential. It is inferred from mobility (movement) measurements in an electrical or acoustical field with a bunch of assumptions (which may or may not be valid). Thus the optical properties of the particle are not required. A Google Scholar search will show papers indicating where the electrophoretic mobility has been used to determine the zeta potential of bacteria.
