I measured Na in my plant extract (shoots or roots ash+water+ HNO3 and HCl) 3 months a go and now that I repeat the measurement with the same samples the absorption are more than 10 times higher. Did you have such an experience? What can be the reason?

Would you please explain your sample extract preparation in detail?
Did you use old plant extract after three months or use dried root/shoot and again you prepare new extract\?

بنده هم دليل نكته ايي كه در خصوص افزايش جذب نوشته بوديد برخورد نكرده ام.فقط دو سوال داشتم
اول اينكه آيا شما همان محلول استخراج شده قبلي را سنجش كرديد يا اينكه دو باره از همان بافت خشك اولي استخراج مجددا و سنجش داشتيد
دوم اينكه در صورتي كه به جواب سوالتان رسيديد لطفا براي بنده هم نتيجه را ارسال نماييد چون من هم سنجش دارم. اما روشي كه من استفاده مي كنم هضم بافت خشك در اسيد نيتريك نيم مولار براي 48 ساعت مي باشد و از اسيد كلريك استفاده نمي شود.

This is the protocol I used:
1.2 g of seeds was dried at 80 ˚C for 48 h and then 1 g of dried seeds was ashed at 550 ˚C for 12 h. The furnace used for ashing was Ceramic Engineering Furnace Manufacturers, Sydney, Australia. It was adjusted on a rate of 30˚C/min to reach 550 ˚C, kept in 550 ˚C for 10 h and then cooled to 30 ˚C. 6 ml of 10 M chloric acid and 16 M nitric acid (1:4 ratio) (Terfi and Sadi, 2014) were added to the ashes and then made up to 50 ml volume with deionized water. The solutions were subsequently sonicated with the Unisonics, Sydney, Australia for 10 minutes and centrifuged at 4000 rpm for 10 minutes. These solutions were used to read absorption with Atomic Absorption Spectrometer (Varian SpectrAA 220).
I used the same samples (I kept them in plastic containers with no evaporation).
Susan, thanks for your suggestion, but why HCl alone is better than mixture? I saw that HNO3 (with higher molarity) is usually used for metal determination in plant tissues.
