My Supervisor always ask me to keep or maintain the A260 more than 20. Why it is so important? Can we just count it by the concentration of nucleid acid?
A260 is a peak OD for nucleic acid
A280 is a peak OD for protein
A230 is a peak OD for phenol
A260/A280 in the range 1.8-2.0 : The extract is rather clean
A260/A280 less than 1.8 : The extract may be high contamination by proteins and the other substances
What if the range of A260/A280 is more than 2.0?
If you have a OD of ~2, that means you got the best RNA.
A value of 2.1 is considered as pure RNA compared to that of 1.8-2.0

The ratio of optical density at (A260/A280) determined the purity of the extracted nucleic acid . This ratio should be in the range of 1.8-2.
So the ration should not exceed 2.0?
It can be up to 2.1.
