What is the reason for using these salts after elution? I have been using 0.1 M NaCl for purification of protein. If I use NaCl in place of KCl what may be the consequences?
most of the ion exchange matrices contains Na+ or Cl- ions as anion exchanger or cation exchanger. when we eluted our desired protein by using buffer having varied amount of salt or other thing. so we used salt solution of higher ionic strength in comparision to elution buffers for washing. so NaCl is selective salt for matrices washing and doesn't change its properties.
These salts have high ionic strength, and can precipitate proteins bu salting in method. when the sample is treated with NaCl, it will remove all the unwanted resin particles bound to it leaves a comparatively pure compound. same way, it will increase the solubility of proteins, and make high egree of elusion in the elusion column.
