If there is a possibility to cut the sample in half (longitudinally) you could use atomic force microscopy (AFM) to determine the surface roughness of your sample.
Dear singh, you would have to cut your sample as the deon said, and then you can measure roughness by using AFM either in contact mode or tapping mode. you can also measure roughness by using optical based zeta profilometer ( 3-D optical profiler) but in that case also, you would have to cut the sample.
