I gave out some tissue samples to a commercial company for metabolomics analysis using LC-MS/MS. After receiving and analyzing the data, a few metabolites (unsaturated fatty acids) were shown to be differentially unregulated. I now want to validate these metabolites in our lab using some routine lab methods. I'm not such which methods (one that can be carried out in an average BCL1 lab) are suitable for validating unsaturated fatty acids. Good suggestions/protocols/relevant papers etc are welcome and appreciated.

most frequent method for fatty acids is GC-MS, including one-step derivatization into the fatty acid methyl esters (FAME). this approach is used for all kind of biological samples, including tissues, bacteria, plants etc. the derivatization is simple and the analysis is robust. the excellent resolution capacity of the GC-MS helps to separate even unsaturated species.

@Christoph Wittmann. Thanks Christoph for your prompt information. I really do appreciate it. I thought because we used LC-MS/MS in the initial metabolomics screening, an independent assay (not involving chromatography/mass spec) might be required for the validation.
Try to search bioprotocols on google. I have a feeling they have a few protocols.
