I want to culture few cyanobacteria like Anabaena, Nostoc calothrix etc, in large scale for getting biomass.
Can anyone tell me the suitable media for this culture? BG11 media not giving so much biomass. I am culturing in Glass tank of 36"*36"*26"
please give reference also.
Prashant is right. You can use BG- 11 media with nitrogen for better production. Before that you should confirm the purity of the media. Now, when you are starting large scale cultivation you should keep minimum required initial inoculum.
Ideally 10 mg dry weight algae can go in 1 litre of media. Accordingly you calculate for the your tank.
Provide good mixing and aeration to your culture which will help to reduce the desolved oxygen in media, which becomes toxic above optimum levels.
if you are after biomass, then by all means use the full strength medium (I would actually use half strength: with such high concentrations your cultures are not likely to become nutrient-limited. I also always thought that phosphorus in BG11 is low, but then I am a fan of the F medium even for freshwater algae). air bubbling is necessary both to maintain your algae in suspension and to eliminate excess oxygen. pH control would help, but one important detail you do not mention is light:, which might become limiting if lighting is from one side only, since 36" is a fairly long optical path. you might try increasing light intensity, use lighting from both sides or overhead and probably you could use 24 h photoperiod.

hi. here goes Richmond's Manual. It has no recipe for the F medium, but you may find it useful. see the part on optical path, for instance (page 140 and following) and you may find an explanation for poor growth, different from the composition of your growth medium
As to the f medium, here goes a manual, unfortunately in spanish because it was for our own students initially, and for latin america when it as publishe.
I have translated the relevant parts of appendix 1 for you. The recipe is for the half strength, which is probably too low, but the solution is simple: for the full f medium, use 2 mL/L of the stock solution and if you wish to try higher strengths, simply use more stock solution (thus, 4 mL/L will give you a 2f medium, 6 mL a 3f medium etc),
for the growth medium, use filtered, sterilized tap water, dechlorinated if necessary, not distilled or purified. Of course you may also wish to try different media, but i never found a real advantage changing growth medium, compared to the notable improvements derived from e.g. changing the geometry of the container or other conditions. For instance, did you check temperature preference of your species? What about pH? not all blue greens will grow well above pH 9-9.5
