With the same ligand, I have seen antimicrobial activity with some other sets of complexes. E. coli is one species but I can't get the zone of inhibition this time. What may be the problem and why it is happening?

Can you describe your ligand that you used (it's fine if you can't describe the ligand, it may still be possible to make general observations)? It sounds like you saw a zone of inhibition in the presence of some metal-ligand complexes and did not observe the zone in the presence of other metal complexes with the same ligand. Did you see any inhibition with the ligand only? Did you do control experiments where you have the metal present only for comparison? It sounds like there is metal uptake that is occurring in the presence of the ligand, which could result in toxic levels of the metal being taken up by the bacterium, but I'm not certain without answers to the other questions.
