I prepared i.v. injectable nanoparticles. I would like to study the drug release from these colloidal formulations. I can use pH buffer 7.4 but it will be for quality control purposes. To understand the mechanism of drug release (but not really a QC method), I want to use relevant media that simulates physiologic conditions. Will the addition of some amount of BSA work? Please share your thoughts!
I think that the choice of the release medium should be based on the purpose of the prepared nanoparticles, i. e., if nanoparticle are designed for injectable forms, a buffer of pH 7.4 will be suitable.

Yes, the nanoparticles are designed for i.v. injection. pH 7.4 buffer does not have any protein content. I am not sure if simulates physiological conditions! I totally agree that pH 7.4 buffer is optimum for regular QC purposes.

I totally agree with You that the buffer (pH 7.4) does not exactly simulate the physiological conditions, but keep in mind it is an (in vitro) experiment. In addition, the in vitro release medium should not contain any additive that might interfere with the drug.
I also consider that pH 7.4 is well suited for in vitro studeis. But if you the site of action of drug is stomach then pH must be around 1.8 or so, and if the drug has to be delivered to large intestine, it must be studied at pH 8.6 or so. So the action of site is important.
I agree with former answers that your release media depends on your supposing site of action of drug. As for i.v., pH7.4 PBS, 0.1M is commonly accepted as media. Besides, to be more convincing, if you can perform your experiment in flow-through-cell dissolution tester recommend by USP is preferable.
I agree and you want to test the release in physiological close viscosity then 55% serum in PBS will also be sufficient. Let m know if you have any more questions. Best wishes,

Yes, adding appropriate % of BSA to buffer will help mimic the blood plasma. Also, in addition to this, you will come to know if your nanoparticles are interacting with the BSA or not. Best.
The problem is that only BSA is not present in blood plasma but other proteins are also present along with other biomolecules. So, artificially preparing a mimicing release media is rather difficult task as the other entities present will also interfere in in vitro release process. Therefore, PBS is best.
