I was centrifuging 80% amonium sulphate saturation for partial purification of Bacteriocin at a speed of 10000 RPM for 20 mins at 4॰C. But my falcon tubes broke incide the rotary chamber.
please suggest me the preferable centrifugation speed and time.

Without knowing the exact rotor diameter, nobody can tell you, other than to give rough estimates. The relative centrifugal force will vary, and is greater for a large diameter rotor than it is for a small diameter. You should be able to determine the relative centrifugal force in g (force equivalent to the force of gravity) from tables that come with the rotor and/or centrifuge. If not, it can be calculated by the formula RCF=1.18xr(Q/1000)^2 where RCF is relative centrifugal force in "G", r=radius of the rotor in cm, and Q=rpm. To answer your question, we often use about 3000xG with satisfactory results. To prevent tube breakage, make sure your vessels conform to the shape of the rotor well; if they don't, use a soft plastic or rubber cushion adapter. Also, use thick-walled tubes or bottles meant for centrifugation.
