I have 10 ug of IL-4 stock, and I want to dilute and aliquot this stock. what are the solution that I had to use to dilute and aliquot IL-4 stock? PBS, Distilled water, or other?
It is hard to tell without knowing which strain of mice you are using, which dose of virus you gave them and which time-point after infection you are analyzing. In my hands, BAL dilutions can vary from no dilution at all to more than 100x, depending on the experimental setup and cytokine being determined. I would recommend taking few representative samples - one with expected low amount of cytokines (control animals) and one with expected highest amount (infected animals at the peak of the response) and run an ELISA with serial dilutions (2x, 5x, 10x, 20x, 50x, ...) to determine the optimal one for your samples.
