there would be many approaches that you can use for measuring photosynthesis rate. I suggest to use LI-6400XT Portable Photosynthesis System and make the light and CO2 response curves on your plants. In this way you can find which part of the curve influenced according to the experimental set up that you used for your study. If you want to correlate it with the biomass you can use the following formulas:
relative growth rate=net assimilation rate*specific leaf area*leaf weight ratio
net assimilation rate=(1/leaf area)*(dw/dt)
dw/dt would be the difference between two weight in two measuring time
another equipment that you can use for studying photosynthesis can be fluorcam which you can visualize different parameters related to fluorescence of chlorophyll.
Rate of leaf photosynthesis can be measured using LI-COR (LI-6400 Portable Photosynthesis System) on 6 cm2 leaf area at bright sunlight condition and at certain radiation level with an adjusted cooling temperature of 25 °C. The radiation level will be determined after measuring the prevailing radiation level of the experimental area. Good luck
