A GC-FID or GC-MS approach would be quite easy if you have the tools available. A quick pubmed search for cholesterol by GC-FID would get you a simple protocol that you could work up.

I am sure it can be done with HPLC but I am not confident as to what detector you would need or deritizations that might need to be carried out first.
Yes, I am sure too, but the problem is related to the cost of the pre-treatment. There are many papers in which researchers detected the presence of cholesterol through many pre-built kit, but it is much costly method.
