I would like to consider multiculturalism as an independent variable in a multivariate regression to figure out it’s effect on quality of learning in a big city. Can you advise a good measure way for multiculturalism?

I would suggest you track down aspects of the MCP indicator that relate (or can be translated in)to an educational setting, and use this as your Independent Variable.
How are you defining "multiculturalism"? It's a term that requires qualification. Check-out Goldberg (1994) Multiculturalism. A Critical Reader. It will get you started.
It depends on the context in which you want to measure multiculturalism. Is it in the school setting for example in which case you would need to check the curriculum but also the basic principles and measures taken at school level. Likewise, if you want to check the broader societal level, are we talking about civic engagement in which case you would need to check the level of inclusion of different minorities in civic action, citizenship and other such issues. It is a very broad which in many cases links also to radicalization and hate speech but of course this depends on the focus of your research.
I would consider to analyze statistics: either ethnic groups and educative groups (both formal and non-formal education). And to contact with associations that work with those city ethnic groups

From my practitioner experience, I would conduct a community mapping assessment to understand the degree of multi-culturalism and pluralism that exists in a given community using a structured interviewing tool as well as conducting focus groups with a random sampling of a target community. However, we must first build awareness and educate targeted communities on working definitions of multi-culturalism and pluralism to generate buy-in for these community mapping methodologies.
there is a need to go back and define your term, "multiculturalism" with details of exactly what you are looking at. one important reason for this is, there may be demographic diversity with much segregation. i have lived in a two large cities where this is the case-- many ethnic groups but little or no interaction amongst them.
