There are various methods for the determination of ABA. The one which we use,is solvent extraction, partitioning and then HPLC determination. extract fresh or freeze dried plant material with 80% methanol at 4C for 48 h, in darkness.the extract is evaporated under vacuum to aqueous phase. make the aqueous phase to 50 ml with distilled water, and make pH 2.5 using dilute HCl. then , do partitioning with diethyl ether four time . combine the ether phase and evaporate at 35-40C to 2ml. then use HPLC with detection at 254 nm, using internal standard.
