I want to use the Hydroxyl Radical Scavenging method for a series of antioxidant tests.
In the written protocol, I should use iron sulfate, salicylic acid and 0.1% hydrogen peroxide.
Does 0.1% hydrogen peroxide mean by weight or by volume?
and What is the solvent of ferrous sulfate in this protocol? Is the Distilled Water?

0.1% H2O2 simply means 0.1ml. H2O2 in 100ml. distilled water
FeSO4 soln.generally means water as a solvent. PBS also can be used based on the sample to be assayed.

Solvent is dilute water. So the percentage refers to percentage volume of hydrogen peroxide in diluted by distilled water.
Hydrogen peroxide is available as 30% solution (very dangerous!) or 3% solution. It has to be appropriately diluted with distilled water.

thank you for posting this interesting technical question. I think you have already received a sufficient number of expert answers that your question is fully answered. I would just like to add a useful suggestion to the point "Does 0.1% hydrogen peroxide mean by weight or by volume?" When you have a 3% commercially available solution of hydrogen perpxide available and want to make a 0.1% solution, it is perfectly okay to work with volumes, as the difference in density can be neglected. For this and other related dilution preparations I suggest to use what we call in Germany the "mix cross" (see attached picture). Supposed you want to prepare a 0.1% solution from a 3% solution and a 0% solution (water), you write the starting concentrations on the left side of the cross as shown and the target concentration in the center of the cross. On the right hand side you write the differences (3 - 0.1 = 2.9 and 0.1 - 0 = 0.1). The result is that you just need to take 0.1 ml of your 3% H2O2 solution and dilute it with 2.9 ml water (or multiples thereof). 😎
