High power Flame torch is used for Pyrex tube sealing.
for freeze pump thaw cycles you can use a rubber septum to close the pyrex tube temporarily.
In my opinion, Glass ampoules or Biotage microwave vials are more convenient to use for COF synthesis instead of Pyrex tubes.
