If u want to germinate it in soil then soak the seeds in water for atleast 12 hrs, the soil must containing half sand and half clay mix with FYM...try it .....i hv already done by this method....it wrks. the geminatin will be observed after 7 days.
Sir can you please tell me ....why this is good time to germinate the tobacco seed...........is thr any specific reason or use?...........please let me know..........
you may be knowing that particular plants will grow in particular season...like in kharif and Rabi season..because of physiological maturity....flowering and seed set etc...........
Dear all you cant take seed germination on agar. If you take it out it will die and it can be suggested only for academic purpose and not practicable. pls use mixed media of coir pith (cocopeat) and sand in 1:1 proportion (field capacity) and put seeds on it you will get very good percent germination
for germinating any seed, best approach is on agar based media. make simple 0.6% water agar on petripltes.
Then sterilize the seeds with 0.1% HgCl2 and ethenol. Time will be vary for different seeds. then soak the sterilized seeds in autoclaved distill water for 1-7 days depend on thickness of seed coat. After that dry these sterilized seeds on autoclaved blotting paper. All these will be done in laminar airflow. Then put the seeds in petriplates and keep the plated in dark for 2 days followed by light. By doing a little manipulation in this method u can germinate any type of seed in any season.
I have old tobacco seeds & want to germinate......... What are you meaning by old? in what conditions were they stored prior to the idea to germinate them .... 30 years ago when dealing with Germplasm I discovered in Litterature that in USA they have a collection that is tested every 25years for viability the last 200 years...... and vitality was 90% plus.I used seeds in a petrie dish with watersoaked filterpaper and a fungicide. Sometimes had to rescue the seedlings witha bacteriacide. Then with needle transplanted the seedligs (3 days old) to a clay potting mixture on which I had drawn a lattice design with the needle before taking the seedlings to the intersects and just put a thin layer of the potting soil to the roots .... 4 weeks later I transplanted the noww the 6 to 10 cm seedlings to a fibrepot nursery befor planting them in the nursery for seed rejuvination for the germplasm collection. May you have success....... it was nice seeing very old varieties e.g from 1938 when the Tobacco Germplasm collection was started.
I tried agar innitially but the "dirt" on the seedcoats was growing faster than the tobacco and I ended up with the most delightful coloured collections of fungal and bacterial colonies around the tobacco seeds. The closed petri dishes were put in an incubator (darK) at 24-28 degrees Celsius..... and watered daily as the incubator dries tha filterpaper which capilarry......

Well you should follow the normal aseptic seed germination protocol. Use one 100 micron mesh.
Clean the surface by hypochlorite treatment. keep the seeds in 0.1% HGCl2 for 5 mins. wash thrice in sterile D.H2O. spread them over 1/2 MS 0.8% agar plates. cover with foil and keep at 25+- 2 c.
