The supernatant equals the theoretical total with nanaoprecipitation method for preparation gatifloxacin nanoparticles with this formula.
The organic phase: aceton 5ml +PCL 200mg
the aqueous phase: buffer solution 20ml + drug 20mg + PVA 1.5%
acetone is evaporated with rotavapor and the nano suspension is analysed for encapsulation efficiency... and the result was so negative...where is the problem?
By reducing the concentration of PVA, encapsulation efficiency can be improved. even i had a same problem while formulating microparticles.
It's better to use W1/O/W2 double emulsion method. Since gatifloxacin solubility is pH-dependant, you can make the inner water phase (W1) slightly acidic, while making the pH of the external phase (W2) slightly basic to make it unfavorable for the drug to escape. The method you're using (nanoprecipitation) may not be suitable for hydrophilic drugs.
By the way, it's also possible to formulate nanoparticles with nanoprecitation method without the use of a surfactant. You need to optimize the conditions first. But as I said, it's not in general recommended for hydrophilic drugs.
