I want to measure stomatal size (length) in order to correlate it to ploidy. Now I remember from plant physiology classes long ago, that stomata become shorter and wider when they open. It seems therefore, that the momentanous condition of the plant matters when the imprint is made.
In the methods section of taxonomic literature I haven't seen any mention of this, sofar.
Is it important to measure stomatal size exclusively on open (or closed) stomata, and if so, which condition is prefered?
