All the solutions given by the software in the optimization part estimates a lower yield than one of the experimental runs (#22). I think it is better to take run #22 as the optimized condition because I found it reproducible and the production is quite good. But then, why doesn't the software include this as one of the solutions for optimization?
Note: one of the factors in run #22 falls in +a value
This is a ccd experiment, intended for medium optimization. So i chose the range based on previous report and made the reported optimal condition as my centre point!
Before Doing any RSM experiments; It is preferable to do One factor at a time based optimisation. The reports from previous experiments may not be as accurate for your design......
The minimum and maximum ranges is critical for getting a better RSM design...
The bioprocess variables perhaps behaves crazy..
so now can i use run #22 as the optimal point or is it a MUST to rerun the whole experiment with corrected ranges?
The Design predicted the solutions based on algorithms and they tried to minimise the errors...The actual values may not be same as the predicted values....
If you can reproduce the run#22, you can take it as the optimal points....
Did you run all the solutions and got almost similar values to the predicted values given by the software??
When i set the range of parameters at -1 to +1, yes the real values were close to the predicted ones. Run #22 is reproducible too. But when i set the range at -a to +a, i get much lower values than what is estimated. But this is explainable in biological terms. I did not run all the solutions anyway.. Just took the best 3.
Will it give rise to negative comments in my article if i report conditions for run #22 as optimal point?!
