Does anyone know if there is any RNA-seq cDNA library kits that can generate both the cDNA library for sequencing and for qRT-PCR for gene expression assay (for seq result validation)?
To my knowledge, RNA-seq cDNA lib. prep. usually involves RNA fragmentation before reverse transcription. Therefore, these cDNA are not suitable for gene expression qRT-PCR anymore.
I through that most people would take some of the RNA (from some tube of RNA for seq lib. prep.) to do a normal RT, getting cDNA for qPCR. However, my boss think that two RT reactions (RNA-seq cDNA lib. prep vs normal RT) generate cDNA pools with disparity. Likely due to differences in enzyme, RT conditions, RT primers, etc.... Therefore, we are looking for RNA-seq cDNA library kits that generate cDNA that is suitable for both purposes.
Or do you have alternative ways to minimize/ eliminate the cDNA pools disparity?
