I am shopping around for cheap but yet efficient transfection agents for primary cortical and hippocampal culture neurons. I have been using Lonza's nucleofactor and the efficiency is high (40%). Downside, need high number of cells per transfection, expensive and electroporate before plating. With all the vendors telling me how great their new product is, anybody can recommend any other reagents that is worth my time or money? Try modified calcium phosphate (clontech) protocol before, was not too happy with the health nor the efficiency.
I am going to try the Mirus version of nucleofactor replacement and Ozbioscience Neuromag magnetic transfection system hopefully soon. I will post my results up if people are interested.
The Mirus version of nucleofactor works pretty well. Very comparable to Nucleofactor with a little higher toxicity but difference is minor. Just plate more cells. Neuromag works but gets 2-3 cells per coverslip, similar to Lipofectamine.
