I have fairly small amounts (+- 20ml) of plant crude extracts (methanolic).
I was wondering what would be the best technic to use for a first fractionation.
First liquid liquid or column?
What are the pros and cons?
I'm trying to have a phenolic fraction and a terpenic fraction.
If column, what stationary phase? And which solvents?

i think liquid liquid extraction should be first as column chromatography is cumbersome process
If you have 20 ml methanolic crude extract, firstly you should remove methanol and freeze dry it to get solid dried crude extract. Then you can go for fractionation either by column chromatography or flash chromatography. Flash chromatography will be better, later for isolation work, you can use column chromatography. Choice is as per your type of work.
