Hello Everyone; I am working in the isolation of Hofbauer cells from Placental tissue, using percoll gradients after physical mincing the tissue and sequential collagenase digestions for 15 minutes. I have no experience with percoll and would like to ask if anyone does have with the isolation of these cells using pecoll. I tried gradients of 50%-40%-30% and 20% centrifuged at 1000g x 20 minutes but no clear band is seen. I would ,kindly appreciate your input and experience. Would ficoll work?
