I need to fatty acid profile analysis (FAME preparation and GC) of seaweed (Ulva and Glacilariasp) and green algae also. I tried to extract the oil by Bligh Dyer method. However, the oil percentage of above type of sample was low and difficult to separate it. Can you give the suitable separation method for oil extraction for above samples?
I often use hexane-acetone extraction of fresh biomass (homogenized and sonicated) and add water to the extract until an unpolar upper phase seperates containing strongly non-polar compounds, followed by TMSH derivatization to get the FAME. On occasion, centrifugation is needed to facilitate phase separation. Use of internal standards is strongly recommended. Anyway, no experience with seaweed.
