My Qubit readings are sometimes out of range (too high). When this happens I dilute my RNA 1:5 in RNAse free water and I get values that are too low, like 115 ng/ µL. If I multiply 115 x 5, then I should have gotten a concentration within range in the first place.

Dear Daniela, it is correct to dilute the samples. Due to the detection range (I believe The assay kit is designed to be accurate for RNA sample concentrations between 250 pg/µL and 100 ng/µL) your original readings cannot be accurate. Maybe do some more dilutions and check if these are in line.

Boris Schmitz , the assay range of my kit (RNA BR assay ) is between 20 and 1000 ng/µL.

If so, you need to check RNA quality/ purity. Something disturbes your measurement.
