I am trying to isolate miRNA from postmortem brain material to use in OpenArray analysis. I am using miRVANA kit and I did a first trial with about 50 mg of tissue, and 600 ul of Lysis buffer, the rest of the protocol was as detailed by Ambion, and at the end the nanodrop gave me a value of 260/280 higher than 2; however the 260/230 ratio is of 0.27, is this normal for miRNA isolation? Does someone have experience with this kit? The centrifugation times were pretty short (about 10 secs) so I do not know if they are enough. Any advice would be greatly appreciated.
