Silica get column chromatography is very useful even in isolating the pure molecules. As you are focusing on the semi-purified fractions it very much doable.  

For me it looks you are doing a bioactive fractionation of extract for that you can simply fractionate you extract with different polarity of solvents (of choloroform:methanol or any other suitable solvent mixture)  loading on a silica column.

As far as getting semipurified fraction of a crude extract is concerned, silica gel column chromatography is good enough.

Silica gel column chromatography is a good method to get semi-purified fraction or even pure compounds, what you need is the good eluent for getting good separation

All colleagues that wrote before me are right, just an additional comment: Test CH2CL2-MeOH systems or EtAcO:MeOH by TLC, before you start by traditonal silica or flash system.

Extract the bark first with 70 % then fractional separation on silica gel using methanol:chloroform: few drops of water polarity increase step wise until you reach 100 % and few drops of water 

Yes you could, however, first you should examine the proper solvent system you have to use in column. This examination could be doable by developing your extract on TLC. Keep in mind that it is not necessary to find what is good in TLC will be good in Column. It just a good indication to know how to  proceed then properly. What is more is the volume of your column. The big, the better separation. Finally, examine your fractions after running your sample on column to see the quality of separation. In the case of finding overloaded bands on TLC, I suggest to do subtractions before going through column. And you can start from this latter point to see how complexity your extract is. Good luck!

if u r in a hurry, VLC is good option, then partitioning, then column chromatography with appropriate solvent system.

since your extract is etoh, Chcl3: meoh will be good option in gradient composition.

of course you can get semi pure fractions. Preferably start with coarse type then go for the fine one in order to purify your compound.

At this time I suppose you have run a Silicagel  CC and you have semi purified fractions, and of course you have tried TLC for each of them.  The question is how much semi purified fractions you need. Semi purified fractions containing 5 spot or 10 or more than ten?.  

it is better to start by defatting your crude extract with n-hexane or pet. ether. then go directly to the VLC using silica gel and gradient elution technique. you will obtain different fractions  in nearly a semi-purified form.

Yes you can!... The slower you change the solvent ratios, the most resolution or separation. All of course also depends on your sample. If it's the ethanolic fraction you want to separate, I would start with Petroleum ether, and start slowly moving to diethyl ether 98:2, 95:5, 9:1... and see how your TLC goes...

At most cases, you can absolutely use silica gel chromatography to get semi-purified fractions. I suggest you to follow that with a rapid method to check whether the purification is sufficient, such as TLC- thin layer chromatography for each fraction (or even HPLC) and hopefully you can see one band (or peak) for each fraction...GOOD LUCK.

Yes it is possible to purify compounds using silica 

But it needs efforts, and sometime it is quiet difficult because of similar polarities

you need to do it many times. (Fractionate fractions until luckily you get pure compounds)

However try to use sephadex LH 20 if possible to fractionate using molecular weight it is appropriate and helpful after vacuum liquid chromatography (silica)

You need to know that maybe using isochratic conditions after solvent fractionation on silica will be better if you choose to work only with this method
